Description
This hypotonic solution very quickly causes complete lysis of the red cells. The hemoglobin being lost the ghosts of the red cells become invisible under low power ( they could be seen under high power ). Though the W. B. C.s also lose their cytoplasm, they show their nuclei prominently - fixed by the acid and stained by the gentian violet. The number of white cells is counted in a relatively large amount of space and under the low power of the microscope.
